Sinopse:
O game se inicia logo após os eventos do primeiro game, Desmond Miles consegue escapar da Abstergo ( que foi revelado ser uma organização de templários modernos ), com a ajuda de Lucy Stillman, que revela ser uma assassina. os dois vão para um esconderijo, onde Desmond conhece Rebecca Crane e Shaun Hastings, que construíram uma nova versão do Animus, conhecida como Animus 2.0, Desmond então vê as memórias de um novo assassino.
Já no Animus, Desmond vai até o século XV, na Itália, durante a Renascença, onde viverá as memórias de Ezio Auditore da Firenze, um assassino que incialmente busca vingança pela morte de seu pai, mas após fugir de Florença ( sua cidade natal ) com sua mãe e sua irmã e encontrar refúgio na Vila Auditore, Ezio descobre que seu pai era membro da Ordem dos Assassinos. sendo assim, Ezio passa a ajudar seu tio Mario na eterna guerra entre Assassinos e Templários.
